Pharmacological Properties

 Pharmacodynamic Properties:

Doxycycline is a bacteriostatic antibacterial that acts by interfering with bacterial protein synthesis in sensitive species. Doxycycline is a semi-synthetic tetracycline derived from oxytetracycline, which acts on the 30S subunit of the bacterial ribosome, to which it binds reversibly, blocking the union of the aminoacyl – tRNA (transfer RNA) on the complex formed by mRNA and ribosomes, preventing the addition of new amino acids onto the growing peptide chain and interfering with protein synthesis Doxycycline has a broad spectrum of activity (gram-positive and gram-negative bacteria). It is active against Mycoplasma hyopneumoniae Bordetella bronchiseptica Pasteurella multocida. There are at least two mechanisms of resistance to tetracyclines. The most important mechanism is due to the decrease in the cellular accumulation of the drug, caused by the establishment of an elimination route by pumping the antibacterial or as a result of an alteration in the transport system, which limits the absorption of tetracycline dependent on Energy. The alteration in the transport system is caused by inducible proteins encoded by plasmids and transposons. The other mechanism is due to the decreased affinity of ribosomes to the tetracycline-Mg complex.2+, caused by ribosomal mutations.

Pharmacokinetic data:

Bioavailability after oral and IM administration is very high. When administered orally, values exceed 70% in many species. Food may slightly modify the bioavailability of doxycycline. Under fasting conditions the drug may have approximately 10-15% higher bioavailability than when fed to the animal. Doxycycline is widely distributed in the body thanks to its physical-chemical characteristics, since it is highly fat-soluble. It reaches both well-irrigated and peripheral tissues. It is concentrated in the liver, kidney, bones and intestines, because it has an enterohepatic cycle. In the lung it reaches higher concentrations than in plasma. Therapeutic concentrations have been found in aqueous humor, myocardium, reproductive tissues, brain and mammary gland. Plasma protein binding is 90-92%. Approximately 40% of the drug is extensively metabolized and excreted in the feces (bile and intestine), most of it as microbiologically inactivated conjugates.
