Melanin transporter Kinesin

Kinesins, a type of transporter protein, pick up melanosomes enclosing tyrosinase and melanin, and transport them toward the dendritic tips of melanocytes along path called microtubules. Melanosomes are then delivered to epidermal cells, which darken them.Suppression of kinesins stops the transport of melanin to epidermal cells and eventually lightens the skin tone. Since CinderellaCare has been confirmed to have activities to effectively suppress the expression of kinesins, it is expected to perform skin-lightening effects.

Suppression of melanin migration within melanocytes

Melanosomes are usually synthesized at the center of melanocytes and transported to dendritic tips by kinesins. When CinderellaCare were added to cultured melanocytes, the kinesin activity were suppressed and primary melanosome ( or externally added pseudo melanin) were not carried. As a result, melanosome remained around the center of the cells.

Whitening effect in 3D skin model

Each well of 3D skin model containing melanocytes was exposed to 20 mJ/cm2 of UV-B ray irradiation. Then, 35 μL of each sample was applied on the stratum corneum layer. UV-B ray was irradiated and the samples were added every other day and the cells were cultivated for ten days in total. As a result, CinderellaCare showed a concentration-dependent effect to suppress melanin production which was higher than arbutin and tranexamic acid. Thus, CinderellaCare is expected to perform skin-lightening
effects.

Pigment Spots are Decreased

A lotion containing 1 % CinderellaCare and a control were applied on one side each of the face of 21 male and female test subjects in their 20’s to 40’s for 2 weeks and the darkness of the skin (melanin index) was observed and the number of dark spots on their face was counted. In the area that CinderellaCare was applied, the melanin index significantly lowered and the number of dark spots reduced as compared to before the test.
