    CLOCK® Laboratory Study

    CLOCK® laboratory work showed significantly positive effects on circadian clock proteins. CLOCK® demonstrated to modulate the expression of circadian clock proteins in C6 animal brain glioma and IPEC-1 cells. Oxidative stress can cause problems with circadian rhythm dysfunction. This research showed that CLOCK® reversed the damaging effects of oxidative stress on key proteins related to circadian rhythms. CLOCK® was also shown to stimulate BDNF.

    In House Pilot Study

     

    After the laboratory work, an In House Pilot Study was performed having subjects fill out a questionnaire. Subjects consumed either 500 mg or 1000 mg of CLOCK® an hour prior to bedtime and improvements were observed. Some felt they could sleep through the whole night compared to their previous sleep patterns of waking up and tossing and turning through the night. One subject taking 1000 mg felt a major difference in sleep quality after day 3. Whereas, others felt improvements the first night. The results of this trial led to the first clinical study.

    Two Double Blind Placebo Controlled Studies

    In 2016, CLOCK® was studied with 32 subjects, in a double-blind, placebo-controlled, randomized human clinical study. The results showed that CLOCK® at a 1 gram dose significantly increased plasma levels of BDNF, a protein that is key to brain health and alertness. In addition, there was a significant increase in irisin and melatonin levels and an increase in acetylcholine (Ach) levels. Subjects showed an improvement in attentiveness, alertness, focus, feeling energetic, enthusiasm, and feeling well-rested with a reduction in feeling sluggish and feeling depressed without the same improvement in the placebo group.

    The second CLOCK® study in 2017, was double-blind, placebo-controlled, randomized, and a crossover design with 30 subjects. They were considered to have impaired wellbeing, low energy, and sleep problems. The results of the study showed that CLOCK® significantly improved the important biomarkers BDNF, irisin, acetylcholine (Ach), choline, and melatonin.

    Research

    Research shows that a person who has a variation in the gene of BDNF may have an increased risk of obesity. A study developed by the National Institutes of Health (NIH) was performed that showed a particular difference in the BDNF gene that may influence some individuals to become obese. These particular individuals have lower levels of BDNF produced, which is considered to be a regulator of appetite. The investigators of the study stated that it would be therapeutic to increase the BDNF levels of these individuals.

    Brain-derived neurotrophic factor (BDNF) - It is an important protein that influences brain function and the central and peripheral nervous systems. It is involved in neuron development and repair. Elevated levels have been shown to improve mood and support cognitive function.  BDNF also plays a role in exercise capacity and mitochondrial function in skeletal muscle. Research has demonstrated that BDNF increases with aerobic exercise in different portions of the brain. In the clinical study, those taking CLOCK® had greater attention and focus, which confirms previous research done on BDNF. The study also showed that CLOCK® increased plasma BDNF levels similar to what is seen in acute strenuous exercise. So while you sleep you get the same cognitive benefits with increased BDNF, as if you exercised!

    Acetylcholine (Ach) - It is a brain neurotransmitter critically important in cognitive function, sleep, and muscle control. It is highly concentrated in both the peripheral and central nervous systems. Ach declines with age and problems begin to occur in the brain as Ach drops along with cholinergic tone fails also, which is believed to reduce cognitive function.

    • Many products on the market make claims stating they support mental function based on borrowed science and use commodity ingredients. The actual ingredients in the products have not directly demonstrated to increase Ach.
    • They may make claims on commodity ingredients based on other research showing it inhibits acetylcholinesterase, which thereby would assume it increases acetylcholine.

    Choline - It is the precursor, a building block to Ach, and has been studied because of its role in mental function and memory. There is a transporter that carries arterial plasma choline through the blood-brain barrier where neurons have it available for use. Choline is stored in a phospholipid membrane as phosphatidylcholine. It is then made available to manufacture acetylcholine to be used as a neurotransmitter key for attention, alertness, arousal, motivation, and memory. It also activates muscle groups.

    Irisin - It is called the exercise hormone because it increases with physical activity. Elevated levels are associated with helping maintain healthy body weight, improve cognition and slow the aging process; it may predict telomere length in healthy adults, which would be considered to be an anti-aging feature. It may help with metabolic problems related to obesity.

    Melatonin - It is considered the sleep hormone that is secreted by the pineal gland in the brain. The maintenance of balanced levels in the body is important. It helps to maintain circadian rhythms related to the sleep/wake cycle and it helps the regulation of various hormones including female reproductive hormones and their involvement in menstrual cycles.
