Science:
Gravity plays a relevant role in sagginess as it is a constant force pulling downwards on the skin, in addition to several other external factors as well as the natural aging process.
Collagen and elastin work in tandem to provide firmness and tightness to the skin. In addition, other intracellular elements help to reinforce skin cohesion to counteract flaccidity. The synthesis of both proteins decreases with age and they suffer higher degradation from matrix metalloproteinase enzymes, mostly activated by UV radiation. To improve skin structure, it is necessary not only to increase functional elastin synthesis but also its correct assembly, which is modulated by the key proteins FBLN5 and LOXL1. The complex forming elastin fibers with FBLN5 and LOXL1 additionally binds to integrins, which are transmembrane receptors that also connect to focal adhesions, helping to increase cellular adhesion to the ECM.
Uplevity™ peptide boosted FBLN5 and LOXL1 protein levels in vitro, which are key for proper elastin assembly, upregulated the expression of genes involved in collagen synthesis and focal adhesions and induced the synthesis of collagen I and elastin in vitro. In vivo, it helped increase skin firmness, restructured the dermis and offered a tensor effect on the lower face.

Improving Skin Organization:
19 women aged between 50 and 60 years old displaying saggy skin applied a cream containing 2% Uplevity™ peptide solution on one half of the face and a placebo cream on the other half, twice a day for 55 days.
The fragmentation of the upper and deeper reticulum was determined by confocal microscopy and the results were used to calculate the equivalent age of the volunteers’ skin through a linear statistical correlation.
